Capsules are the most-shipped supplement format in the world, and the one where quality differences are hardest to see from the outside. This guide explains how encapsulation actually works, what separates a real manufacturer from a bottler, how US regulation treats these products, and who the top 5 capsule manufacturers are for eCommerce brands.
A capsule supplement is a measured powder fill inside a shell: actives, fillers and flow agents blended to uniformity, then dosed volumetrically into a two-piece gelatin or HPMC capsule. The shell hides the formulation entirely — no taste, no texture, no visual cue — which is precisely why capsule quality has to be proven in data rather than judged by the customer.
Production runs in three technical stages, and a failure in any one of them shows up as an under-dosed capsule, a capsule that will not open, or a lot that fails a marketplace test — never as something a buyer can see.
Step 1
Actives, fillers and flow agents are screened and blended to a validated uniformity. Capsule filling is a volumetric process, so bulk density and flow behavior — not just the recipe — determine whether every capsule carries the same milligrams.
Step 2
Dosator or tamping-pin machines compress the powder into a slug and seat it inside a gelatin or HPMC shell. Machine speed, tamping force and shell moisture all move fill weight, so the line is checked continuously against a weight-variation spec.
Step 3
Capsules are deducted, polished and metal-detected, then counted into bottles with desiccant and induction seals, torque-checked, labeled and lot-coded — with disintegration and dissolution testing confirming the dose actually releases.
Many companies marketed as "capsule manufacturers" are in fact bottlers: they buy bulk finished capsules from a third party and count them into your bottle. That is a legitimate business, but it means the formula is not yours, the excipient ratio cannot be changed, the cost structure is fixed, and any potency question has to travel back up a supply chain you do not control. A true manufacturer owns the weighing, blending and encapsulation — and can therefore change a spec when your product needs it.

Six checks that separate a genuine encapsulator from a company that buys bulk capsules and fills your bottle.
Encapsulation doses by volume while your label declares milligrams. Ask how often capsules are weight-checked on the line and what the accepted variation limit is. A manufacturer who cannot state a number is not controlling your dose.
A capsule that does not open is a capsule that does not work. USP disintegration testing — and dissolution where the active warrants it — is the only proof that the dose is actually released rather than passing through intact.
Bovine gelatin, HPMC, pullulan and enteric shells behave differently with hygroscopic, oily or acid-sensitive actives. The right shell is a formulation decision, not a checkbox — and it decides vegan, Halal, kosher and delayed-release positioning too.

Capsule supplements are regulated in the United States as dietary supplements under the Dietary Supplement Health and Education Act of 1994 (DSHEA). The format carries obligations that are easy to overlook precisely because nothing is visible:
Dietary supplement cGMP requires written specifications, identity testing of every incoming dietary ingredient, batch records and finished-product verification. For capsules it also means documented blend-uniformity and weight-variation procedures — an encapsulation step you cannot verify is a dose you cannot claim.
Disintegration testing to USP standards, and dissolution where the active warrants it, is what demonstrates the capsule opens and the ingredient becomes available. Over-polished or over-dried shells can fail this even when the fill is perfect.
The Supplement Facts panel must state the dietary ingredient, the part of the plant where applicable, and the amount per serving. Proprietary blends may state a total, but the ingredients must still be listed in descending order by weight.
Bovine or porcine gelatin, HPMC, titanium dioxide, magnesium stearate and colorants all belong in Other Ingredients. This is also where vegan, Halal, kosher and allergen positioning is either supported or quietly contradicted.
You can say a product 'helps support joint comfort'; you cannot say it treats arthritis. Disease claims turn a supplement into an unapproved drug, and the FDA disclaimer must appear on the label.
Heavy metals, microbiological limits and pesticide screening apply as they do to any supplement, and botanical identity testing guards against adulteration. Capsules add moisture control, because a shell that absorbs water becomes brittle or sticky — which is why desiccant and induction seals are part of the spec. California Prop 65 thresholds are stricter than federal defaults for any DTC brand shipping nationwide.
